The left side volume toggle buttons for raising and lowering ringer and alerts is not working anymore. Only way to control the ringer and alerts is from inside the settings itself. What to do? iPhone 4s 5.0.1 jailbroke

Restore the phone to make sure it's not a hardware issue. If the buttons work after restore then it was a Cydia package you had installed causing the issue. If they still don't work after the restore then you have a hardware problem.

The buttons actually "work" just sounds can only be adjusted from inside the settings tab. Hopefully software issue
You do have the "Change with buttons" option in Settings > Sounds set to ON don't you?

My iPhone 4 used to do that, hasnt happened on my 4s. The cause was dirt in the charge unit, the phone thought an
accessory was attached. Cleaned it with a toothbrush and it worked again.

Did a fresh restore last night and all is good
