If you update it by having made a custom ipsw and thus maintaining the baseband, then you can re-unlock it. If you update / restore it via itunes by just hitting the update or restore button, then yes you will make it unlockable. If you need help feel free to message me , I will need to know what iphone you have and what ios and what baseband (Settings - general - about - modem firmware) so I can give you instructions.
